Project description:
In our laboratory we perform mass spectrometry-based proteomics, metabolomics and lipidomics analyses across organisms and species and specialize on functional metabolism research. We have developed and applied activity-based and affinity probes based on click chemistry for enzyme discovery and profiling, and several other functional proteomics and metabolomics methods (e.g., one carbon metabolomics, combined redox metabolomics and proteomics) routinely applying state-of-the-art mass spectrometry infrastructure (2 Bruker timsTOF pro/HT) coupled to different chromatographic systems.
The PhD student will develop functional (meta-)proteomics workflows employing activity-based probes for discovery and profiling of different enzyme classes. Sample preparation and LC-MS/MS data analysis and integration will need to be established, adapted and optimized for different sample types (e.g. human, mouse-associated and environmental microbiome samples, microbial (co)cultures). The development and application of functional meta-proteomics and its integration with complementary multi-omics workflows will provide a better mechanistic understanding of host-microbiome and environmental microbiome phenotypes, activities and interactions.
